Penetapan Kadar Tablet Metoklopramid Hidroklorida Secara Ultra Performance Liquid Chromatography (UPLC)
Antiemetics are drugs used in the management of nausea and vomiting. Metoclopramide Hydrochloride is a prokinetic agent used to treat nausea vomiting associated with stomach irritation, gastric static, irregular intestinal obstruction, the effects of chemotherapy drugs, and radiotherapy. This test was carried out with the aim of determining the levels of active substances in metoclopramide hydrochloride tablet preparations in branded generics and logoed generics. The test results are then compared with the requirements referring to the Indonesian Pharmacopoeia edition VI of 2020, namely Metoclopramide Tablets containing C14H22CIN3O2.HCl.H2O, equivalent to not less than 90.0% and not more than 110.0% of the amount stated on the etiquette. The method used in this test uses Ultra Performance Liquid Chromatography (UPLC) with a column of 2.1 mm x 100 mm (3.5 μm) C18 filler material and an isocratic mobile phase in the form of a buffer mixture of Sodium Acetate P : Acetonitrile P : Tetramethylammonium Hydroxide P in Methanol P (1: 5) then set the pH of the solution to 6.50. With a flow rate of 0.2 mL/min, PDA detector. Sample testing was carried out three times with 2 different drug samples. Based on the tests that have been carried out, the average level of metoclopramide hydrochloride in sample P is 94.205% with RSD of 0.53% and the average level in sample G is 91.826% with RSD of 1.46% so that it can be concluded that the sample meets the requirements.
